Neubauer counting chamber is a thick glass slide with a specialized grid used under a microscope to manually count cells in a liquid sample, such as blood or sperm. It has a precisely known volume, allowing for the calculation of cell concentration in a sample by counting cells within a specific area and then extrapolating that number to the total volume. Key features include a grid with different sized squares to count various cell types, a standardized depth of 0.1 mm, and the ability to load two samples independently.   Key features and function Material and construction: It is a thick glass slide, measuring approximately \(30\text{\ mm}\times 70\text{\ mm}\times 4\text{\ mm}\).  Counting grid: The central area features a double grid, and each side has a \(3\text{\ mm}\times 3\text{\ mm}\) counting area.  Grid subdivisions: Each \(3\text{\ mm}\times 3\text{\ mm}\) square is divided into nine large \(1\text{\ mm}\times 1\text{\ mm}\) squares.  The central large square is further subdivided into 25 smaller squares, which are then divided into 16 tiny squares, resulting in a total of 400 small squares within the central square. This area is used for counting red blood cells and platelets.  The four corner large squares are used for counting white blood cells because of their lower concentration, which requires a larger area for a more accurate count.  Standardized depth: The chamber is designed to maintain a precise depth of \(0.1\text{\ mm}\) between the bottom of the grid and the specialized coverslip.  Volume calculation: The standardized depth allows for the calculation of volume. For example, one of the nine large squares represents a volume of \(0.1\text{\ L}\) (\(1\text{\ mm}\times 1\text{\ mm}\times 0.1\text{\ mm}\)).  Methodology: A diluted and well-mixed cell suspension is carefully applied to the chamber.  Using a microscope, cells are counted within a specific, predetermined area of the grid.  The number of cells counted is then used to calculate the total cell concentration in the original sample, using the known volume of the counting area.  Common applications: It is used to count various blood cells (red blood cells, white blood cells, platelets), bacteria, yeast, and other cells in a fluid.
